Dontari Poe was born on August 18, 1990, in Memphis, Tennessee, to black American parents – Robert Poe Sr. and his wife Sandra Poe. He is the youngest of three brothers with his two older brothers named Pierre Poe and Robert Poe Jr. He grew up in the Whitehaven district of Memphis, Tennessee, where he attended Wooddale High School. While practicing with the marching band in Wooddale, he was discovered by his school soccer coach, who later invited him to join the school soccer team.

Dontari played as a defensive tackle and had eight sacks and 63 tackles in his youth, earning him an honorable mention as a national defensive lineman. Outside of school, Poe played for Auburn, Mississippi, and Colorado. He also participated in athletic events. In his junior year, he was the class 3-A shot put champion, and in his senior year, he also retained the title. Because of his athletic ability, he was awarded two stars by Scout.com and Rivals.com.

Dontari enrolled at the University of Memphis where he played college football for the Memphis Tigers. In his freshman year in 2009 and the following year, he had 27 tackles to his name and became the first defender on his team. Poe finished the 2010 season with 41 tackles and received an honorable mention from the All-Conference-USA.

Poe signed an $11 million four-year contract with the Chiefs. During his rookie season, he battled with Jerrell Powe and Anthony Toribio for the starting jersey. After spending three pre-season games on the bench, Poe was given the starting jersey after Toribor was injured. He distinguished himself by becoming a permanent starter for the rest of his rookie year.

He played four seasons for the Chiefs, and when his contract expired in 2016, he moved to the Atlanta Falcons with a one-year $8 million contract. In the 2017 season, Poe recorded 39 combined tackles, including 23 solo and 2.5 sacks in 16 games for the Falcons.
